Back 1. Prepare a temporary mount of root tissue2. Focus an optical microscope on the slide, set the objective lens on the lowest magnification3. Use the coarse adjustment knob to move the lens down above the slide, use fine adjustment knob to re-adjust the focus until the image is clear (select higher magnification if needed)4. Count the total number of cells in the field of view and the number of cells in a stage of mitosis5. Calculate mitotic index
